I read in one of the "best of the best" guides I believe about the 500 mafia member restriction for "normal pvp".
Does this also is true for fighting bosses in the Boss event and the upcoming Syndicate Wars?

Interesting thought that it might not apply in the tournament.... Try it and let us know? Probably the 500 really is the limit. Would be interesting if it were not can you imagine the scramble for more mafia members.
True, thatīs why I am asking this.
Also for Boss events, if the total attack count, and not only the attack for the first 500. Then it would be interesting as well to gather as many mafia members as possible.
But, is there anybody out there that can shed light into this matter?

your a/d is calculated off 500 mafia max. All calculations for wars, PvP, bosses, etc are taken from this number, so having more than 500 mafia doesn't have any benefit. If you look at the restrictions in the main MW event, and remember cc and MW run off the same platform, this had the same 500 ally limit for attacks and there is no reason to expect this to change.

The 500 limit also applies when fighting bosses. According to the spreadsheet, I should be able to kill level 17 bosses with my current stats, and thats exactly where I top out. If my real attack were based on more than 500, I'd be able to kill much higher level bosses.
